An O2-sensing diguanylate cyclase broadly affects the aerobic transcriptome in the phytopathogen Pectobacterium carotovorum

Pectobacterium carotovorum is an important plant pathogen responsible for the destruction of crops through bacterial soft rot, which modulated by oxygen (O 2 ) concentration. A soluble globin coupled sensor protein, Pcc DgcO (also referred to as GCS) one way P. senses oxygen. contains a diguanylate cyclase output domain producing c-di-GMP. Synthesis second messenger c-di-GMP increased upon binding sensory domain. This work seeks understand regulation function at transcript level. RNA sequencing and differential expression analysis revealed that deletion only affects levels in cells grown under aerobic conditions. Differential showed alters metal transporters. These results, followed inductively plasma—mass spectrometry showing decreased concentrations six biologically relevant metals deletion, provide evidence can affect cellular content. findings improve understanding level control O -dependent phenotypes phytopathogen establish basis further studies on c-di-GMP-dependent functions .
